What do you mean by all "missions"? And I already completed the game but there is a "secret spot" on each movie? Do I have to find those?

Hit them four times. The fourth hit won't seem like it connected, but the portrait will become visible above the character's head. If you try to hit them a fifth time, you'll break them and will have to start over.
Alternatively, you can also do it without the "beating up" by taking over a villain character that you previously bought. The portraits automatically appear for those guys. --- CyricZ - So broken

I can find nowhere in the game manual or any hints within the game itself that you have to beat up enemy characters in order to buy them in port.
I don't mind experimentation, but there needs to be some sort of clue given. Hitting a character (four times, no more, no less, with no indication after the first three hits you are making any sort of progress toward a reward) is incredibly random and arbitrary, in my opinion.
I was with the OP, I kept waiting for Blackbeard to "unlock" somehow. Never knowing all I had to do was hit him a few times.
Is there something I missed in game or in manual that tells you this, or is it just something they expect you to "Figure out"? If it's the latter, I'm going to be quite disappointed in TT.
