Well seeing as how Altair lived in the Holy Land he most likely spoke Arabic or Hebrew, however he (and the other assassins) must have learned english seeing as how they could communicate with Robert de Sable and Altair could talk with King Richard.
Ezio obviously spoke Italian but also learned Turkish so he could communicate with Yusuf and the Turkish assassins in Revelations (he even comments that his Turkish sucks when he dresses up as a bard.
AC3 is obviously not going to need many translation explanations seeing as how everyone was English and we will probably see native languages being spoken when Connor talks with natives from his village or other fellow natives in the frontier.

What about Alta´r? I know Ezio talks in Italian for the entirely of ACII, yeah? Haven't played Brotherhood nor Revelations, does Ezio stick to Italian?
And ACIII will likely be fully in English?
-Desmond definitely speaks English the entire time and seems to have no knowledge of other languages.
-Due to Altair's setting, I'm assuming he grew up speaking Hebrew/Arabic, but learned English (or at least understood the language). After all, many of his targets spoke English, as did his wife.
-Ezio speaks Italian for the entirety of his life (a bit of French, as there were some French girls in Firenze; he probably knows some Spanish as well due to his hunt for Cesare Borgia). By Revelations he probably knows a bit of Turkish, having helped build the Turkish Assassins to their glory and all.
-Connor seems to speak English a majority of the time, though also knows his native Mohawk tongue.
