With how cynical this industry is against Nintendo, it's save to say that they'd get a truckload of hate and ridicule if they made a Mario Galaxy 3, no matter how good the game ends up being.
Adding to that, they kinda wrote themselves into a corner with the escalation from land to world to galaxy. Mario is in space now. Where do you go from there?

Here's an idea: Dimension/reality shenanigans.

Have Bowser step on the history eraser button or whatever which messes up reality or fuses together different parallel dimensions in some way. Mario then has to travel through 6-8 screwed up dimensions to collect the missing power stars (for it is always stars) and fix up the fabric of reality.

Part of what made the Galaxy games so good is that there were next to no rules. There was no limit to what they could throw at you in terms of level design because spaaaaaaaaaaaace.
Well, with reality messing up, there's even fewer rules now.

Granted, the Mario series never cared to explain its level design rationally, but the Galaxy motif gave the whole thing some context.
The dimensions / reality thing would allow the same + even more trippy stuff.

I think Nintendo will go ahead with Galaxy 3. The name carries some weight. Combined, the Galaxy games have sold over 17 million and are two of the highest rated games of all time.

A new "Galaxy" would generate some buzz, a lot more than SM3DW did. I think it would be in Nintendo's best interest right now to leverage the Galaxy brand.

And if they made Galaxy 3, they'd have an excuse to release the trilogy together as well.

I don't think it has to keep on getting more macro in scope. They could always just go to something like a city or village, and have Mario jump in various paintings or whatever else they could think of. A bit off-topic, but a franchise like Final Fantasy just keeps getting less and less grounded, with both its themes and its settings. That didn't necessarily make it better than previous entries into the series.

Why not expand on the whole galaxy motif and make it Super Mario Universe?

Because that wouldn't change anything. The individual levels in Galaxy 1 & 2 are Galaxies which span the cosmos. The scale is already as massive as players could reasonably take in.

Careful, remember what happened with Big Rigs: Over the Road Racing and that game's lack of rules.

The trucks could accelerate infinitely in reverse, putting Galaxy-class starships to shame.

Yeah, but if we went with TC's warping dimensions and stuff idea, they could justify it as being loads of different universes, including parallel ones or whatever. I think Universe would probably be a title they'd use.
