Can you have two of your characters in a game at the same time, using the same profile?

No. Only one controller can be assigned to a profile. Only that profile has access to it's characters. You can have a 'guest' sign in on a second controller, but they will only have access to level 1 characters and they(you still will) won't save any progress IIRC.
