I just started NG+ and was going to try no kills. How do you assassinate the REQUIRED marks when you are using the path of silence? Does it allow you to kill them in a different manner or will I have to figure out some way of killing them?

You'll have a sword for the required marks and assassinate them as normal. You pull it out of god knows where.
