What exactly are they referring to when they say "Do not miss any charged shots"?

I have done everything I can and still not earn the mastered achievement.

That means, any charged shots you make, you can't miss any of them. That is, every Charged shot you make must either hit an enemy with it's blast, or it must actually go into the Kraid's open chute (not simply impacting on the side of the chute or something, causing a Miss)

Could I use the regular beam shots (Tapping the ZR button) on the targets, or do I still have to use the charge shots to hit the target signs?

Everything can be destroyed using regular shots. The only *required* charge shots is the chute in the mouth. (Kind of like shooting those torpedoes into that port in the Death Star, really)

You actually don't even need charge shots to the mouth if you play two players, since you can ferry a wii-remote player up into the mouth to shoot Kraid's belly target with basic lasers.

I'm not sure if refusing to use any charge shots would disqualify for the master rank, though.

I found this master rank to be a lot easier to accomplish piloting the Gunship, rather than playing on foot.
Although I don't know if you need to master both kinds of play or just the one.
