Can you download a dlc while playing a game?

If so does the game have to be played off the disc? Any exceptions? I know you can't play on-line while downloading.

I think so. I downloaded GoD Borderlands 2 while playing Lost Odyssey today.

Depends on the game.
Some games utilise a constant Live connection. Gears of War 3, for example, has a drop-in-drop-out Co-op feature which means it's using Live all the time. If the console is using Live, downloads are paused. You'll know because a popup will, erm, pop up and tell you.

Using live and installing a game are the only things that will stop downloads.
