I never played Prototype 1 so shouldn't I enjoy Prototype 2?

It has better graphics, gameplay, missions etc.

I did play Hulk Ultimate destruction and it wasn't my kind of game I found it quite boring after a while.

Prototype 1 had better gameplay than 2 Alex was much more agile and felt smoother to control.

If you didn't like Ultimate Destruction I highly doubt you'd like this series, it is very similar.
