I'm looking for some games that have a co-op campaign or something we would both enjoy. She isn't into games like Call of Duty, and we don't like the multiplayer experience on NSMBW (I'm always having to help her).

It's not co-op since it's a board game, but Fortune Street is really cool, and you can play the both of you against some CPU's (there's always 4 players) and see who's better. I got it REALLY cheap new off Amazon not too long ago (around 15) and it's quite cool.
According to my case, Pikmin 2 has both co-op and versus, but I haven't tried it. I imagine it's pretty cool though, since the single-player part rocks.
Trauma Team and Trauma Center: New Blood both have a 2-player mode, but I haven't tried that either, again, the games on their own as single-player are cool.
And Star Successor: Sin and Punishment is a shooting gallery sorta game from Treasue, which you can play alone or two-player, again, I haven't tried the co-op though.

Go Vacation could work for co-op/multiplayer and the difficulty is very forgiving in most activities. I have tried co-op before, but I don't know if it requires player 1 to unlock everything first/whether it's possible to unlock stuff during co-op.
Ivy the Kiwi? supports multiplayer/co-op. It might be a little too difficult if either of you do not enjoy using the Wii pointer controls.
Otherwise, you are probably looking at games that are primarily single player with the option of a supporting second player, for example: Mario Galaxy series, Rabbids Go Home, Sin & Punishment 2, etc.
