I almost exclusively play Lulu (basically anywhere but jungle). It's a huge advantage to know a character that well over someone who doesn't.
That being said, I played in a few tournaments, and it was inconvenient that my team had to pick around me, and bans/picks were nightmares.
But in all seriousness, when someone plays one character much more than most others, they are probably REALLY GOOD at playing that champ, assuming they have a decent understanding of the game mechanics and solid technical skills.

My main rotates every few weeks. It's just the champion I'll play the MOST during that time period.

Its really context sensitive. If the person literally means that is the only champ they ever play then yeah, they're probably bad. But this isn't Street Fighter, this is League so its unlikely that's what most people mean when they say they main a certain champ.
My preferred role is top lane and my most played champ is Darius. So in that sense, he is my main. I have the most experience with him and I am the most comfortable playing him.
However, if my lane opponent is a particularly difficult matchup for Darius or if my team comp would benefit from me playing a different champ, I can and will play someone else.
Personally I would be happier to see "I main X champion" because I assume that means they will perform well with that champ. Much better than seeing "never played this champ before". --- Kakugo Ha Yoi Ka... Isshun Sengeki! My Fists Bleed Death
