The rifle is for shooting what waits for you at the bottom of the tower (should you choose to use it). If you lose your Angelic Rifle somewhere along the line, you'll get another one before actually getting to the bottom floor. You can also use it on regular enemies, but like you said - there are only 5 bullets.

statusOnce you enter the chamber that god is in, you have the option to shoot god with the rifle, which does the same cut scene each time, or walk into god, which unlocks the next chamber. You need it to get to god, however. The rifle fires one hit kill shots, so use it when you are stuck in a dangerous spot or to kill an annoying enemy. Make sure you always have at least one shot, though.
