I looked on my map to see the galata den is locked, but when I went there I could still go inside and access everything, so I don't know what the difference is.

It means you can't assign anymore recruits in that den.

Means you've got a Master Assassins protecting the den, and Templars won't take it over anymore.

So what happens when you get full templar awareness if you have all dens locked?

if all dens are locked the templars do not attempt to take them. when I got all seven of them locked it automatically completed the defend three dens assassin challenge.

So once all are locked you can get away with buying all shops and not worry about having to do that RTS mini game anymore?

It appears so. I have not had to deal with it since I locked them all. I rarely have full templar awareness for more than about twenty minutes though so I may be incorrect.

Once they are all locked the only outcome is increased agression from guards.
