Do you use these to differentiate between where the game was made or how the game plays?

These terms need to die fast so people can go back to using real sub genre names and actually know what they are talking about.

How the game plays. Obviously how it plays. For instance when I 1st played Dark Souls I did assume it was a 'western' developed game. Now I call it a JRPG but I always point out it's a western style RPG.

JRPGs tend to be more colorful, have simple storys, and worst gameplay. But they have better music, characters (in personality and design) and the atmosphere always seems inspired.

I wouldn't call an RPG Japanese unless it was Japanese.

I usually avoid the terms altogether though because they don't really mean anything useful.

There's a clear and utter difference between a Japanese style RPG and a Western style RPG. Anyone who says otherwise or throws out "egh, u regionally challenge" is a blind fool.
