Is the a way to load up on rupees?

Uh, yes, there are ways of doing so, but it might help if you could say what point in the game you're at.

@DragonEleven To heal yourself, simply sit on the stool for a few seconds (instead of wasting time on grass and monsters)

If you perform well in the Lightning Round mini-game (after a story-related event at Lanayru Gorge), it's possible to get up to 3000 Rupees at once (though this is very time consuming - a full run takes 45 minutes). Perfecting the Dodoh's High Dive mini-game (after getting the party wheel back) gets 500 Rupees - 20 Rupees per play. Mr. Tubert's Thrill Digger mini-game is luck-based, so you might not want to do it, but the hardest difficulty has multiple Red and Silver Rupees, with the occasional Gold Rupee.

There are Goddess Chests that contain silver and gold Rupees, there are items that allow you to find Rupees more easily, Goddess Walls and digging spots can yield money, certain treasures can be sold to Rupin for 100 Rupees each, Sand Cicadas can be sold to Stritch for 50 apiece (with the others generally yielding 10-40), there are some silver Rupees to be found atop pillars in a certain area in Lanayru region...
Most of what's available to you depends on how far you are in the game.

What I found to be a fairly easy way to earn rupees was the minesweeper-like digging game on the volcano (need to bomb a wall to reach it)... it's fairly easy to make your money back and after that you can make a fairly decent amount once you get the hang of it... only downside is that you take damage when you lose (will probably need a bit of luck to avoid that) but you can just go off to heal and then come back.

Go to bamboo slicing mini-game. You need to pay 10 rupees to play, but if you cut between 15 and 20 times, you get 30 rupees, so you have 20 rupees profit every game (less then 15 gives nothing, more then 20 gives treasure)

Not sure if you have the Rupee Medal yet, but that would help if you brought it along on a quest. Earlier on in the game for me, I would aquire quite a number of rupees then end of spending them on adventure pouch slots or something equally expensive, and finding myself with almost no rupees again. Just keep playing, slash every bush or plot of grass, discover chests, sell things if you want (though it's hard to make a LOT of money this way, depends on what items you have/need), and find a good spot that has a lot of grass or whatever that is also near a door to that area, so you can collect, leave, then come back so everything is respawned. This can work well for bugs and birds too

Play pumpkin pull,if youre good with the bow he charges 20 and gives 50 if you get 150-190 points maybe you can go higher but i think if u get 200 u get treasure.

you can either play on the colorful island or the rupee digging game in Eldin volcano. once you start losing hp in the digging game just go to the stool to heal up

After you obtain the goddess harp there should be several places where there are swarms of blessed butterflies if you play the harp the goddess symbol will appear on a wall. Ready your skyward strike and then draw on the wall a picture of a rupee and then a cluster of rupees will appear, usually a red rupee and a couple of green and blue rupees. (Note: the effects of the wall will regenerate over time and also you can use the walls for bombs, arrows, fairies, and hearts) (Goddess walls are mainly in temples but some are outside)

The goddess walls regenerate when you save and quit. Find one near a save point, and save/quit over and over drawing rupees on the wall.
